Differential membrane binding of α/β-peptide foldamers: implications for cellular delivery and mitochondrial targeting

چکیده

The intrinsic pathway of apoptosis is regulated by the Bcl-2 family proteins. Inhibition anti-apoptotic members represents a strategy to induce apoptotic cell death in cancer cells. We have measured membrane binding properties series peptides, including modified α/β-peptides, designed exhibit enhanced permeability allow entry and improved access for engagement members. peptide cargo based on pro-apoptotic protein Bim, which interacts with all proteins initiate apoptosis. α/β-peptides contained cyclic β-amino acid residues increase their stability permeability. Dual polarisation interferometry was used study each two different model systems mimic either plasma or outer mitochondrial membrane. impact structure also investigated, results demonstrated that peptides had increased affinity significantly altered bilayer. showed presence an RRR motif ability bind insert into mimic, provide insights role selective targeting peptides.

Peptide-mediated cellular delivery.

Peptide-mediated molecular therapeutic delivery systems have recently emerged as an alternative means to effectively substitute or augment present gene therapy technologies, e.g., TAT, VP22, engineered peptides. These systems show great promise for the elimination of the main bottleneck to safe, efficient, targeted gene therapy delivery and are able to efficiently introduce DNA, antisense pepti...

Peptide-based carbon nanotubes for mitochondrial targeting.

In the present study, we report the design and synthesis of peptide-based-multi-walled carbon nanotubes (MWCNTs) to target mitochondria. Targeting these intracellular organelles might open the way to develop alternative systems to address diseases related to genetic mutations in mitochondrial (mt)-DNA, by delivering therapeutic oligonucleotides. The first step towards mitochondrial delivery of ...

Hyaluronan-binding peptide for targeting peritoneal carcinomatosis

Peritoneal carcinomatosis results from dissemination of solid tumors in the peritoneal cavity, and is a common site of metastasis in patients with carcinomas of gastrointestinal or gynecological origin. Peritoneal carcinomatosis treatment is challenging as poorly vascularized, disseminated peritoneal micro-tumors are shielded from systemic anticancer drugs and drive tumor regrowth. Here, we des...

Maximizing gene delivery efficiencies of cationic helical polypeptides via balanced membrane penetration and cellular targeting.

The application of non-viral gene delivery vectors is often accompanied with the poor correlation between transfection efficiency and the safety profiles of vectors. Vectors with high transfection efficiencies often suffer from high toxicities, making it unlikely to improve their efficiencies by increasing the DNA dosage. In the current study, we developed a ternary complex system which consist...
